Resumen en portugués brasileño: Uma fertilização nitrogenada adequada pode reduzir os efeitos deletérios do estresse salino e promover o crescimento vegetal, sendo esse efeito dependente da fonte de nitrogênio (N) utilizada. Objetivou-se, assim, estudar a influência da fonte de N na tolerância de plantas de girassol (Helianthus annuus L.) à salinidade. Para isso, sementes de girassol foram semeadas em vermiculita, em casa de vegetação, e após oito dias, as plântulas foram transferidas para um meio hidropônico com N sob as formas de NO3-, NO3-/NH4+ (3:1) ou NH4+. Decorridos seis dias, adicionou-se NaCl até se atingir a concentração de 75 mM. As plantas foram coletadas aos 7 e 14 dias após a imposição do estresse salino. O crescimento das plantas de girassol foi reduzido pela salinidade, contudo as plantas nutridas com a mistura NO3-/NH4+ apresentaram maior crescimento da parte aérea, nas condições salinas. A condutância estomática (gs), a transpiração (E), a fotossíntese líquida (A), a concentração interna de CO2 (Ci), a eficiência instantânea de carboxilação (A/Ci) e a eficiência do uso da água (EUA) foram pouco influenciadas pela salinidade, entretanto as plantas nutridas com NH4+ apresentaram os menores valores de gs, A, A/Ci e EUA, em comparação às demais fontes de N. Semelhantemente, as plantas nutridas com NH4+ apresentaram os menores teores de clorofila (a, b e total), de carotenoides e antocianinas, os quais foram pouco influenciados pela salinidade. O vazamento de eletrólitos (VE) e a peroxidação dos lipídios não foram influenciados pelas fontes de N, e a salinidade aumentou o VE. O estresse salino aumentou os teores de Na+ e reduziu os de K+, principalmente nas raízes, contudo esses teores foram menores nas raízes das plantas nutridas com NH4+, em comparação aos demais tratamentos de N. Em condições salinas, os maiores incrementos nos teores de Cl- foram observados nas plantas cultivadas com NO3- e com NO3-/NH4+, entretanto as plantas nutridas com NH4+ foram as que mais acumularam Cl– em seus tecidos. Os teores de NO3- nas diversas partes das plantas e a sua translocação para a parte aérea foram reduzidos pela salinidade, sendo maiores nas plantas nutridas com NO3-, seguidas das nutridas com NO3-/NH4+. Os teores de NH4+ e a sua translocação para a parte aérea foram maiores nas plantas nutridas com esse íon, as quais acumularam também mais N-aminossolúveis e aminoácidos livres, contudo isso não foi capaz de reduzir os níveis de NH4+ em seus tecidos, justificando os sintomas de toxidez observados nessas plantas. Plantas nutridas com NO3-/NH4+, em comparação às nutridas com NO3-, acumularam mais N-aminossolúveis e os aminoácidos glutamina, asparagina, serina e prolina em condições de salinidade. De maneira geral, os teores de N-total, carboidratos solúveis e amido foram maiores nas plantas nutridas com NH4+, não sendo observados efeitos da salinidade nessas duas últimas variáveis. As enzimas do metabolismo do N foram afetadas pela salinidade, independentemente da fonte de N empregada. Nas folhas, as atividades da redutase do nitrato e da redutase do nitrito foram maiores nas plantas tratadas com NO3- e com a mistura NO3-/ NH4+. Houve baixa correlação entre a atividade da sintetase da glutamina e da sintase do glutamato e os teores desses aminoácidos. Esses resultados sugerem que plantas de girassol são sensíveis ao NH4+, apresentando sintomas de toxidez sob essa nutrição, mesmo em condições controle. A nutrição combinada de N (NO3-/NH4+) foi a mais eficiente em reduzir os efeitos deletérios da salinidade em plantas de girassol, pois promoveu maior crescimento e melhor balanço iônico em condições de estresse, além de as plantas sob essa nutrição terem sido as mais eficientes na utilização do N sob estresse.
Abstract: Adequate nitrogen fertilization can reduce the deleterious effects of saline stress and promote plant growth, being this effect dependent on the nitrogen (N) source used. In this experiment, we aimed to study the influence of the N source on the tolerance of sunflower (Helianthus annuus L.) plants to salinity. In a greenhouse, sunflower seeds were sown in vermiculite, and after eight days, the seedlings were transferred to a hydroponic medium with either NO3− or NH4+ as the only N source, or a NO3-/NH4+ mixture (3:1, v/v). After six days, NaCl was added in the nutrient solution until a concentration of 75 mM was reached. The plants were harvested at 7 and 14 days after the imposition of saline stress. The growth of sunflower plants was reduced by salinity; however the plants supplied with the NO3-/NH4+ mixture showed the higher shoot growth under salt conditions. The stomatal conductance (gs), transpiration (E), net photosynthetic rate (A), internal CO2 concentration (Ci), instantaneous carboxylation efficiency (A/Ci) and water use efficiency (WUE) were little influenced by salinity. The NH4+ plants showed the lowest values of gs, A, A/Ci and WUE, in comparison to the other N sources. Similarly, plants supplied with NH4+ showed the lowest contents of chlorophyll (a, b and total), carotenoids and anthocyanins, which were little influenced by salinity. Electrolyte leakage (LV) and lipid peroxidation were not influenced by N sources, and salinity increased LV. Salt stress increased Na+ content and reduced K+ content, especially in the roots, but these contents were lower in the plants supplied with NH4+ compared to the other N treatments. In saline conditions, the highest increase in Cl- content was observed in plants supplied with NO3- and NO3-/NH4+, however plants supplied with NH4+ were the more accumulated Cl in their tissues. The NO3- content in the different parts of the plants and their translocation to the shoot were reduced by the salinity, being higher in the plants supplied with NO3-, followed by those supplied with NO3-/NH4+. The content of NH4+ and their translocation to the shoot were higher in the plants supplied with this ion, which also accumulated more N-amino compounds and free amino acids, but this was not able to reduce the NH4+ content in their tissues, justifying the toxicity observed in these plants. Plants supplied with NO3-/NH4+, as compared to those supplied with NO3-, accumulated more N-amino compounds and glutamine, asparagine, serine and proline under salt conditions. In general, the contents of N-total, soluble carbohydrates and starch were higher in plants supplied with NH4+, and no salt effects were observed in the latter two variables. Enzymes of N metabolism were affected by salinity, regardless of the N source used. In the leaves, nitrite reductase and nitrite reductase activities were higher in plants supplied with NO3- and NO3-/NH4+. There was a low correlation between the activity of glutamine synthetase and glutamate synthase and the content of glutamine and glutamate. These results suggest that sunflower plants are sensitive to NH4+, showing toxicity symptoms under this nutrition, even under control conditions. The combined nutrition of N (NO3-/NH4+) was the most efficient in reducing the deleterious effects of salinity in sunflower plants, as it promoted higher growth and a better ionic balance in stress conditions. Besides the plants under this nutrition were the more efficient in the use of N under salt stress.
