Abstract: Cutaneous leishmaniasis represents one of the main public health problems in our country, along with other endemic diseases such as Chagas disease, schistosomiasis and malaria. Much has been done to assess the extent of the problem, seek solutions to limit it, understand its pathogenesis and establish safe therapeutic approaches. This is the constant concern of the Ministry of Health, through SUCAM. The National Council for Scientific and Technological Development (CNPq) has also been encouraging the study of these problems by financing several projects, and this thesis is precisely the result of this realistic policy that aims to create conditions for a greater understanding of the various facets of the problems and for establishing the best strategies for resolution. The World Health Organization, in turn, has promoted meetings in various parts of the world with the same objective, such as the one recently held in Brasilia from July 10 to 13, 1979, with the aim of critically evaluating the various therapeutic alternatives in use in Cutaneous Leishmaniasis and discussing new perspectives in this field. The histopathological study of cutaneous leishmaniasis emerged at the same time as the identification of the disease in Brazil. Since then, several authors have contributed significantly to the clarification of histopathological changes in skin and mucous lesions, establishing a morphological pattern of the disease that is still used today. This pattern has not been modified even with new immunological knowledge, nor has it been confirmed with a greater number of cases and in different regions of the country, in addition to the Northeast and Southeast regions, where most of the studies were carried out. In view of this situation, we decided to study Cutaneous Leishmaniasis using a large number of cases from two distinct regions - the Amazon Region and the Central-West Region - and to seek a correlation between the clinical and histopathological findings and to establish comparisons between the cases studied from the two regions. With this study, we intend to provide some support for the histopathological interpretation of leishmaniasis lesions. We intend to raise controversial points that cannot be answered by analyzing the slides using routine staining methods, so that in a later study, with the help of more sophisticated techniques, we can confirm the propositions raised here.

 The maximuni values o-F total proteins and lipids&#xD;
 were verifyed in the crude extract obtained in the 28th day&#xD;
 o-F growth and maximum values of carbohidrats were observed&#xD;
 in the extracts o-F the ioth, 22nd and 28th. The fractionated&#xD;
 proteins by SDS-PAGE presented bands with molecular weights&#xD;
 ü-F approximately between i4,4 to 67,0 KDa. The extracts&#xD;
 obtained on the 4th and i0th days presented two bands of&#xD;
 approxini.a.tely ó&lt;S and 67 KDa, the ones obtained on the i6th&#xD;
 and ££nd days, 5 bands of appruximatela i8,5, 30,0; 36,0;&#xD;
 ó&lt;&amp;,0 and 67 KDa and the ones •From the 28th day, 7 bands, the&#xD;
 previouü unes and two more with molecular weights of&#xD;
 approximately 14,4 and 20,i KDa. The carbohidrats and&#xD;
 dininoacids were characterized by Thin-Layer Cromatography,&#xD;
 ctiid presented stains corresponding to sugars glucose,&#xD;
 aã l ac t use and mannose, and the aminoacids asparagine,&#xD;
 pfolirit, thyrosina, al an ine, leucine, isoleucine,&#xD;
 f&amp;'nilalanine, •v'aline, inethionine, trytophan, aicid slutamic,&#xD;
 glycine, histidine e cystine. The antigenicity o-F the crude&#xD;
 t-KÍrací.s was veti-Faecl by Itumunocl i-Pusion reaction in agar&#xD;
 against i-abbifc hiperimmune sera. Precipitation lines were;&#xD;
 observed in all studied extracts and common antigenic&#xD;
 molecular populations. Finally, the allergenity o-F crude&#xD;
 exfcracL obtained art the 28th day o-f growth was con-Firmed by&#xD;
 lyE antibody pruductioit in isogenic mice immunized by the&#xD;
 saine, and titled by Passive Cutaneous Anaphylaxis test in&#xD;
 rat's. The maximum titles obtained were 80 and i60 in cAF-i&#xD;
 «lice- and i.280 and 64^ in BALB/c mice.

Abstract: Thirty male Uistar rats ueighing an average of 50 g uere allocated into three groups of 10 animais: group DT - thiamine-deficient, uas fed a thiarnine-f ree diet and uater ad libitum; group CP - pair-fed control, uas pair-fed uith group DT and received a diet uith all nutrients and uater; group AL - ad libitum control, uas fed a diet uith all nutrients and uater ad libitum. They uere ueighed each tu o. days and the consumption diet and uater recorded. After 19 days on test (beginning of the body ueight loss of group DT) the animais uere sacrificed and small pieces of proximal jejunum uere removed and prepared to study the epithelium through microdissection and parafin inclusion techniques. Before sacrifice the animais uere administered vincristina sulphate i.p. to evaluate the crypt cell production rate. The height of the enterocyte from the middle one- third of villi, the depth of crypts, the length of villi and the cellular populations of crypts and villi uere determined.&#xD;
The results shoued that:&#xD;
1.&#xD;
The animais of the DT group shoued adipsia from 13th day and anorexia from 15th day of the experiment period;&#xD;
2.&#xD;
The animais of the DT group started&#xD;
significant body ueight loss from 17th day of the experiment group in comparison to CP and AL;&#xD;
3.&#xD;
The height of the enterocytes from the middle one-third of villi, the depth of crypts, the length of villi and the celular populations of crypts and villi from DT group uere significantly decreased as compared to CP group;&#xD;
4.&#xD;
The number of crypts/villus and the crypt cell production rate uere similar uhen comparing DT and CP groups.&#xD;
In summary, rats fed thiamine free diet developed initially adipsia followed by anorexia and finally body ueight loss. The adipsia and the epithelial atrophy observed in deficient rats could be responsible, at least in part, for deficient absorption of nutrients commonly observed in thiamine deprivation. Houever, even with epithelial atrophy, the crypt cell production rate uas maintained in thiamine deficient animais, probably due to the presence of immature villus cells, luminal nutrition and adequate DNA production,,
